Introduction

Our team aimed to develop a DNA-based biosensor that could detect eDNA from the microcystin-producing cyanobacteria M. aeruginosa. To implement this sensor, we used Acinetobacter baylyi (ADP1) as our chassis organism due to its natural competency. To engineer ADP1, we used a variety of common molecular biology techniques, as well as procedures unique to ADP1. The experiments outlined below describe how to efficiently engineer ADP1 and can be used as a guide for future iGEM teams.
