We introduce gene fragment which could degrade n-alkanes from Rhodococcus. The main cds of gene fragment alkane 1-monooxygenase gene(alkB) is included, but we also construct it with specific promotor and terminator. We also introduce whole alkB gene cluster for further comparison.
The E. coli-Streptomyces shuttle plasmid is the plasmid template mentioned above.We designed two cloning methods - traditional enzymatic digestion and Golden Gate assembly. The plasmids will be transformed into Streptomyces by the method of conjugal transfer.
We not only use experiment to address the problem but also consider to change the condition from a broad perspective. investigations are conducted to understand the situation of usage of plastic mulch in rural China.
We include two models in our project. We collect data about the usage of plastic mulch at different area of China, and then design a learning model witch can help to find out the rational option for using different mulch at different area. Meanwhile, we also use AlphaFlod to build a model which can help us optimize our target gene sequence.
We hope by this project, more people will be impressed by science. In this case, we set up a club at our high school to share our project and ideas. Through this, we hope more our classmates can have interest in science topic.
