LOGIC (Lasting Optimized Genetic Input Calculator)
Biological computing holds great promise for the future, yet past engineered bacterial circuits for logic gates have suffered from excessive complexity, cumbersome operation, and reliance on multiple input substances. Our LOGIC project overcomes these problems by developing a novel approach to constructing a biocomputer—utilizing quorum sensing and spatial diffusion to form logic gates. For addition calculation, we construct high-pass and band-pass genetic circuits based on the quorum sensing (QS) system. Through the diffusion of QS molecules in solid media, we can build various logic gates and connect them to perform multi-bit binary calculations. For repetitive operation, we engineer a photo-induced QS molecule degradation enzyme by predicting, simulating and adding domains to refresh the calculator. Based on the calculator, we can also build accessories like result storage, decimal result display and latches, demonstrating its extensive prospects. Furthermore, we construct a LOGIC Toolkit with a visualization software for future researches.
