Pre-Lab: Pipetting practice. DNS reagent: 0.5 g DNS, 0.8 g NaOH, 15 g sodium potassium tartrate, water to 50 mL. Standard curve: 6 D-galacturonic acid standards (stock) of varying concentrations. Pectin assay: 8 tubes (4 plasmid / 4 no plasmid): 4.5 mL 1% pectin + 0.5 mL supernatant; incubate 37 °C; collect 1 mL; DNS assay. |

Fruit peels: Prepare peels (Korean pear, watermelon, banana, kiwi, tomato); grind with 50 mL water; centrifuge. Enzyme reaction: For each fruit: 300 µL extract + 200 µL plasmid solution (+/−); vortex. DNS tubes: 50 µL DNS + 50 µL sample; heat 100 °C, 5 min. Absorbance: 80 µL per well in 96-well; DNS-only control; read 540 nm. |
