Basic Parts

This year’s William_and_Mary iGEM team contributed eight basic parts to the iGEM Parts Registry, including one promoter, five coding sequences and two homologous regions. All parts were modified to remove illegal restriction enzyme cut sites, ensuring full compatibility with iGEM type IIS Assembly. These components are assembled into vectors which are used to control biofilm regulation and extracellular protein secretion in Bacillus subtilis and Mycobacterium smegmatis. Bacillus subtilis SinI coding region Twin of BBa_K3498007 & BBa_K4769201 CDS A gene in Bacillus Subtilis that codes for an antagonist to the DNA binding protein SinR which inhibits biofilm coding gene epsA-O and tasA-sipW-tapA.
AmyE integration region - Left arm Twin of BBa_K4103018 Homologous Region A homology arm for chromosomal integration at the amyE locus of Bacillus subtilis, used for stable insertion of genetic constructs via double-crossover recombination
AmyE integration region - Right arm BBa_25Z8K8MR Homologous Region A homology arm for chromosomal integration at the amyE locus of Bacillus subtilis, used for stable insertion of genetic constructs via double-crossover recombination
MSMEG_6752 BBa_25ONKG6K CDS A gene codes for a cellulase enzyme endoglucanase A that breaks down cellulose in Mycobacterium smegmatis.

Composite Parts

We contributed six composite parts to the iGEM Registry, including four regulatory modules that produce key biofilm genes in Bacillus subtilis and one in Mycobacterium smegmatis, as well as an empty integration vector backbone that enables stable chromosomal insertion in B. subtilis. The vector serves as a tool for future Bacillus engineering projects and is fully compatible with both iGEM Type IIS (RFC 1000) and BioBrick (RFC 10) standards. Together, these parts are designed for precise control of biofilm formation and surface protein expression.
