Detective engineering bacteria: Let Pseudomonas aeruginosa have no place to hide in water

Escherichia coli was genetically engineered to construct a dual-modal biosensor responsive to quorum sensing signaling molecules. The engineered bacteria carry a Pseudomonas aeruginosa-specific promoter that drives the expression of β-galactosidase and β-glucuronidase, which hydrolyze the substrates ONPG (o-nitrophenyl-β-D-galactopyranoside) to produce a yellow color and MUG (4-methylumbelliferyl-β-D-glucuronide) to emit fluorescence, respectively. This enables the qualitative and quantitative detection of Pseudomonas aeruginosa.
