Our team designed and built new biological parts to support our project Living Scaffolds.
Among these parts is the pmurI plasmid, a composite part designed to provide a functional copy of the murI gene. This plasmid with the murI CDS allows complementation of delta murI strains. It serves as a tool to validate that our auxotrophic strain regains the ability to grow in the absence of DL-glutamate.
All parts were submitted to the iGEM Registry following the BioBrick assembly standard.
For more detailed information like the sequence, construction details and characterization results, we recommend visiting the individual Registry pages or the Result and Design.
| Name | Type | Code | Description | Function | Result |
|---|---|---|---|---|---|
| murI CDS | Basic | BBa_25ZE7I6P | Coding sequence of glutamate racemase (murI) | Provides the genetic sequence for murI; can be used to construct expression plasmids or other synthetic biology tools | Sequencing of the murI CDS verified that the sequences match the intended design |
| pmurI | Composite | BBa_256Q13UK | Plasmid carrying promoter BBa_J23119, RBS BBa_J428038, murI CDS, and terminator BBa_J428092 in the plasmid BBa_J428341 assembled using Golden Gate. | Expresses murI in ΔmurI strains to restore growth without DL-glutamate; used to validate gene function | Western blot analysis confirmed the expression of MurI protein in our construct, showing a band at the expected molecular weight. Sequencing of the pmurI plasmid verified that the sequences match the intended design |
