Cell-Free Biosynthesis
Cell-free biosynthesis enables the efficient and flexible production of diverse biomolecules, demonstrating immense potential in the field of industrial biosynthesis.
However, the high cost of ATP supply and poor energy stability represent one of the core challenges limiting its large-scale application.
Relevant studies have shown that phosphate accumulation, pH fluctuations, and protease interference all inhibit the activity of ATP regenerating enzymes, potentially leading to a 50%–80% decline in yields during prolonged reactions.
To address this,
we introduce the V-CHARGEs platform.
We used the polyphosphate kinase SIPPК, which exhibits high activity at the optimal temperature and can use low-cost polyphosphate (PolyP) as a substrate.
We encapsulated SIPPК with the porous P22-VLP capsid, which can effectively protect and maintain the activity of SIPPК while enabling the cyclic regeneration of ATP.
V-CHARGEs represents an innovation in enzyme engineering
within traditional cell-free biosynthesis.
