The iGEM Guelph wet lab team has provided optimized protocols for Golden Gate assembly using the MoClo-YTK throughout the design, building, and testing of the various parts of our regulatory genetic circuit. This protocol is useful for iGEM teams and labs that look to complete Golden Gate assembly using the New England Biolabs BsaI-HFv2 restriction enzyme and HI-T4 DNA ligase. Additionally, we have also laid the groundwork and begun the assembly of a library of modular parts that may be used for future testing of modified regulatory circuits that can be designed to detect a variety of ligands and output unique responses. The iGEM Guelph dry lab team has developed detailed protocols
and documentation for the design, fabrication, and testing of
the filter medium containment system. These protocols outline
standardized methods for preparing, freeze-drying, and rehydrating
agarose-based scaffolds, incorporating nutrient gradients and zeolite
additives, and assessing permeability, yeast adhesion, and biofilm
formation. Additionally, the team provides step-by-step guides for
constructing and testing a 3D-printed containment chamber optimized
for water filtration experiments. These resources offer valuable
reference for future iGEM teams interested in developing
biofiltration projects that use genetically modified organisms
