In order to systematically address the limitations of traditional allergen immunotherapy (AIT), such as long treatment duration, safety risks, and complex administration methods. We modified common allergens (pollen allergens, dust mite allergens, and seafood allergens) to obtain hypoallergenic derivatives, in order to minimize the risk of allergic reactions while maintaining T-cell reactivity. During this process, we also constructed both hypoallergenic derivatives and natural allergens. After successful verification, we combined the hypoallergenic derivatives and connected self-assembling peptides (EFK8) to promote oligomerization, enhance stability, prolong the serum half-life, and achieve targeted effects against two allergens in a single molecule.

We have documented these parts and conducted comprehensive tests, including plasmid construction, expression strain construction, protein expression and purification, and functional validation. Our goal is to obtain a safer, more effective, and widely applicable immunotherapy strategy.
