AromaWell – A Customized Aromatic Essential Oil Solution Based on a Programmable Microbial Synthesis Platform

1. Project Overview: Reshaping the "Personalized" Future of the Essential Oil Industry Through Biosynthesis 

 

In the fast-paced modern life, physical and mental exhaustion as well as low mood have become common issues. Essential oil aromatherapy is not merely a sensory experience of scent, but also a gentle and effective way of life therapy. 

The AromaWell project originates from addressing the core pain points of the essential oil industry. Literatures clearly indicate that traditional essential oil extraction is restricted by three factors: plant variety, local climate, and extraction process, making it difficult to flexibly adjust the proportion of components. Moreover, the three core components of rose essential oil (core ingredients: geraniol and citronellol, known as the "Queen of Flowers") and lavender essential oil (core ingredient: linalool, known as the "All-round Guardian") share highly similar terpenoid biosynthetic pathways. 

Based on this key scientific discovery, we propose an innovative solution: directionally modify microbial cell factories (Saccharomyces cerevisiae and Escherichia coli) to build a programmable biosynthesis platform. By deeply integrating synthetic biology and metabolic engineering, end-users can dynamically adjust the synthesis rate of each component according to personal preferences, and finally obtain personalized mixed essential oil components through a single fermentation system. This solution breaks the industry inertia of "large-scale standardized production", enables the realization of personalized needs for "embracing oneself gently with natural scents", and avoids the limitations of traditional processes.

2. Problem Insight: Targeting the "Uncontrollable" Pain Points of the Traditional Essential Oil Industry 

 

The starting point of entrepreneurship lies in accurately addressing unmet needs. Literatures have clearly defined three fatal bottlenecks in traditional essential oil production, which also serve as the cornerstone of the commercial value of the AromaWell project:  


2.1 Passive Restriction in Raw Material Supply 

 

Core essential oils such as rose and lavender rely on the cultivation of specific plants. The local climate (e.g., drought, heavy rain) and variety characteristics directly affect the yield and components, resulting in unstable raw material supply and failure to ensure continuous production. 

 

2.2 Inability to Regulate Component Proportions 

 

Modern users' demand for essential oils has shifted from "general-purpose" to "customized" (e.g., high-proportion lavender for sleep aid, high-proportion rose for soothing emotions). However, traditional extraction processes can only produce products with fixed component proportions, which fail to meet personalized needs. 

 

2.3 High Loss and High Cost in Production Process 

 

Traditional processes require multiple extractions and physical mixing, which easily lead to the loss of component activity. In addition, the high costs of planting and extraction links increase the price of end products and restrict market popularization.

3. Innovative Technical Solution: Building the Core of Biomanufacturing with "Programmability + Precise Regulation" 

 

The technical solution of AromaWell is not a theoretical concept, but a fully implementable innovative technology project based on a mature three-layer architecture of "microbial chassis modification + metabolic network optimization + regulatory system design": 

 

3.1 Bottom Layer: Selection and Modification of Highly Adaptable Microbial Chassis 

 

Two industrial-grade microorganisms are selected as the chassis to balance "metabolic efficiency" and "safety": 

    • Saccharomyces cerevisiae: Complies with the "Generally Recognized as Safe (GRAS)" standard and is suitable for use in cosmetics and aromatherapy raw materials. The focus of modification is to "enhance precursor supply" to provide sufficient raw materials for the synthesis of geraniol and citronellol. 

    • Escherichia coli: The focus of modification is to "introduce selective catalytic enzymes" (such as the GES gene from roses and the LIS gene from lavender) for the directional synthesis of citronellol and linalool, thereby increasing the yield of target components. 

 

3.2 Middle Layer: Core Design of Regulatable Metabolic Network 

 

The key breakthrough of the project is the introduction of a regulatable promoter system and artificial genetic circuits. By adjusting external induction (e.g., inducer concentration) and culture parameters (e.g., temperature, pH), the flux of the respective synthesis branches of rose components and lavender components can be precisely controlled. For example, if a user prefers a "7:3 rose-lavender ratio", the activity of the promoter can be regulated to increase the synthesis rate of geraniol and citronellol while controlling the synthesis rhythm of linalool, realizing the dynamic adaptation of component proportions. 

 

3.3 Upper Layer: Future Direction of Intelligent Optimization 

 

To further lower the user operation threshold, the project plans to integrate "machine learning algorithms and real-time metabolic flux analysis": Users only need to input their preferred aroma ratio of rose to lavender (e.g., 7:3 or 5:5), and the system can automatically optimize the culture parameters and induction parameters, accurately distributing the carbon flow to the target products. This realizes a closed loop of "preference input → parameter optimization → customized output", enabling personalized customization to move from "technically feasible" to "user-friendly".

4. Social and Industrial Value: From "Cost Reduction & Efficiency Enhancement" to "Driving Transformation" 

 

The value of AromaWell extends beyond technological innovation; it lies more in creating sustainable commercial and social value for the industry, which fully aligns with the core requirement of iGEM entrepreneurship—"creating tangible impact": 

 

4.1 Reduce Active Component Loss and Improve Product Quality 

 

By "obtaining mixed components through a single fermentation process", it avoids the loss of component activity caused by "multiple extractions + physical mixing" in traditional processes, ensuring more stable therapeutic effects of essential oils. 

 

4.2 Significantly Lower Production Costs

 

Microbial fermentation does not rely on large-scale plant cultivation, avoiding fluctuations in raw material costs caused by climate and origin. At the same time, it simplifies the production process and reduces the total cost from the source. 

 

4.3 Expand the Boundaries of Industry Product Categories 

 

The project plans to extend its technology to other aromatic combinations with shared metabolic pathways, such as "Citrus-Sandalwood" and "Jasmine- Yilan". This breaks the limitations of existing essential oil categories and creates new product directions for the industry. 

 

4.4 Drive Sustainable Transformation of the Industry 

 

Shifting from "relying on agricultural extraction" to "sustainable biomanufacturing" reduces the consumption of arable land and water resources by agricultural cultivation, which is in line with the global concepts of "carbon neutrality" and green manufacturing.

5. Future Plans: From "Technology Implementation" to "Industry Standards" 

 

The project’s future plans focus on three core directions—"technology deepening, product category expansion, and industry empowerment"—reflecting a long-term business vision:  

 

5.1 Technology Dimension 

 

Prioritize the integration of "machine learning and real-time metabolic flux analysis" to build a dynamic mapping model of "user preferences → fermentation parameters", making personalized customization more efficient and accurate.

 

5.2 Product Category Dimension 

 

After verifying and implementing the rose-lavender solution, gradually expand to other aromatic combinations such as Citrus-Sandalwood and Jasmine-Yilan, and build a product matrix of "full-category personalized essential oils". 

 

5.3 Industry Dimension 

 

Promote the standard-setting for "biosynthetic essential oils", providing industry-referenced norms in terms of component purity, production processes, and environmental protection indicators. This will help the entire essential oil industry transform from "traditional agriculture-driven" to "biomanufacturing-driven".

6. Conclusion: Why Does AromaWell Align with the Entrepreneurship Award? 

 

The AromaWell project fully meets the core requirements of the iGEM Entrepreneurship Award for "innovation, implement ability, and value": 

 

6.1 Accurate Demand Insight 


Clearly identify the three major pain points in the traditional essential oil industry, and address the core contradiction between users’ "personalized needs" and the industry’s "standardized production". 

 

6.2 Implementable Technological Innovation 

 

Based on microbial chassis modification and combined with a regulatable promoter system, the technical solution is scientifically sound and has the potential for lab-scale testing and large-scale production—it is not an unrealistic concept. 

 

6.3 Sustainable Commercial Value 

 

By reducing costs, improving quality, and expanding product categories, it not only meets user needs but also creates new profit growth points for the industry, while promoting green manufacturing. 

 

6.4 Driving Force for Industry Transformation 

 

It goes beyond "developing a single product"; instead, it is committed to changing the production paradigm of the essential oil industry through technological innovation—shifting from "agricultural dependence" to "biomanufacturing"—and possesses long-term industry influence. 

We believe that AromaWell is not just a technical project, but a practice of solving industry problems and creating multi-dimensional value with an entrepreneurial mindset—which is exactly the core connotation advocated by the iGEM Entrepreneurship Award.
