Parts

During our project we designed 10 new parts for comparing IFs variants to find structures suitable for improved B12. Additionally, we designed 2 new parts for expressing cubilin domains 5-8, with the original plan of assessing IF-cubilin binding. Here we present a summary of the designed parts, whereas the full description for each part is documented in the iGEM parts registry.

The designed parts can be divided into two categories: basic and composite parts for expressing IFs and basic and composite parts for expressing cubilin domains 5-8. The parts for expressing IFs have been optimized for K. phaffii and the parts for expressing cubilin domains 5-8 have been optimized for E.coli. The parts for expressing IFs were designed for human, porcine, rat bovine and platypus IF. However, due to patent restrictions ( EP1846769B1) on porcine IF sequence, we chose not to upload the sequence to the iGEM Parts Registry. In our work, the sequence was used for research purposes only and all the results are documented in the Experiments and Measurements sections of our wiki.
